REGULAR BUS : PUNTA ARENAS / PUERTO NATALES ( 3 hrs o/w , 250 kms ) ALL YEAR
There are 3 companies running : Buses Fernandez, Buses Pacheco y Bus Sur. They have 6 or 7 departures a day
From Punta Arenas : 08:00 / 09:00 / 13:00 / 14:30 / 17:00 / 18:30 / 20:00 ( this is Buses Fernandez )
From Puerto Natales : 07:15 / 09:00 / 13:00 / 14:30 / 17:00 / 18:30 / 20:00
At Punta Arenas airport, 20 kms out of the city, there are some transfers to downtown running according to flight schedules. In the city, there is not a main city bus terminal, each company has its own office. You should allow some extra time to get a bus ticket in town, specially in high season.

You get 4 companies doing the service. In low season ( Sep-Oct-Nov / Mar-Apr-May ) there is one departure a day Schedule :

IN : Puerto Natales 07:30 / Laguna Amarga 10:00 / Pudeto 11:00 / Visitors Centre 12:00 OUT : Visitors Centre 14:00 / Pudeto 15:00 / Laguna Amarga 16:00 / arrive to Puerto Natales 18:30 In high season ( Dec -Jan - Feb ) there is a second departure in the afternoon : IN : Puerto Natales 14:00 / Laguna Amarga 16:30 / Pudeto 17:30 / Visitors Centre 18:30 OUT : Visitors Centre 19:00 / Pudeto 20:00 / Laguna Amarga 20:30 / arrive to Puerto Natales 22:30
Make sure to coordinate with the other schedules at the Park services to connect with the Regular Bus. At the Park, make sure you will have enough time if you are doing some trekking.
WINTER TIME : It runs the Full Day Tour Bus, which provides a combination of daily regular bus
JUN - AUG
REGULAR BUS : PUERTO NATALES TO CALAFATE ( Arg ) ( 4.5 hrs, 280 kms ) SEP - MAY
There 2 companies, each company runs every other day. It goes one way one day and comes back the next day. Puerto Natales : Departs at 09:00 / Calafate ( Arg ) : Arrives at 12:30 hrs. ( same hours, the return from Calafate ) You should consider one night stay in Puerto Natales for this combination. ( Check for latest schedules )

REGULAR BUS : PUERTO MONTT TO PUNTA ARENAS ( 32 hrs, 2.000 kms ) ALL YEAR
Normally it would be taken after visiting the Lake Region, then from Puerto Montt to Punta Arenas. This route goes crossing over the Andes and through Argentinean territory on the Atlantic coast, by the cities of Bariloche ( at the Andes foothills ), Comodoro Rivadavia and Rio Gallegos ( both ocean side ). Departures three times a week at 09:30 from Punta Arenas. There are 3 companies running this service.
REGULAR BUS : PUERTO NATALES TO USHUAIA ( Arg ) ( 14 hrs, 825 kms ) NOV - MAR
Bus Sur with departures at 07:30 Monday, Wednesday and Friday from Puerto Natales. They go by Punta Arenas, cross the Magellan Strait at its west end ( at Punta Delgada, by the Atlantic ) on a Ferry ( 20 min ) and go down across the Tierra del Fuego island, by the city of Río Grande and then to Ushuaia. ( Check for new departures )
CAR RENTAL ALL YEAR
You could rent a car, light pick up or a 4x4 Station Wagon, at Puerto Montt, Punta Arenas or Puerto Natales, and do most of this routes all by yourself. It can give you the freedom of scheduling and going wherever and whenever.
